Description

Pasireotide aspartic salt was synthesized by peptide solid phase synthesis method. First, the hydroxyl group of the tyrosine of the peptide chain to be synthesized was covalently bonded with the polystyrene resin (solid phase carrier), and then it was used as the amino component to remove the amino protective group and react with the excess activated carboxyl component to lengthen the peptide chain. Then it undergoes condensation, washing, deprotection, washing, and the next round of condensation with lysine, tryptophan, phenylalanine, (4R)-4-Chemicalbook [(2-aminoethyl) carbamoylxy] -L-proline, and phenylalanine in turn to achieve the length of the peptide chain to be synthesized. The peptide chain was cleaved off the resin, followed by a ring closing reaction in the presence of trifluoro-acetic acid, then the protective group was removed, and finally salted with aspartic acid to obtain pasireotide bispartic acid.
